I know it's the easiest and most obvious thing to check, but check to make
sure the lens is clean and free of dust. I've had what I thought was a very
clean machine, but in one pass it picked up paper dust and lint which
affected the results on the very next sheet.

Just saw this post today... sorry!
My DTP41 has always been very fussy about proper strip length beyond
the test patches. Make sure there's enough material left, out to the
ends of the strips as set up in the page size dialogue.
Other than that, if you can't get a calibration strip read, the device
won't be operationally reliable. It'll need to go back for repair /
recalibration.
You say you're a newbie? Did you purchase the device as rebuilt or
used? What's the date on the supplied calibration strip? That would
indicate the last time the device was put back to factory specs.
