I have installed the Xquartz-2.3.1. I got to 10.5.5 by installing
10.5.0 from DVD, then using Software update to get to 1) 10.5.4 then
2) 10.5.5 this AM. What worked before no longer works. I do not set
the DISPLAY variable. It is currently set to /tmp/launch-CmrQkT/:0. I
do have a .profile defined, linked to .bashrc via a symbolic link.
iTerm and Terminal both hang when trying to source the .profile (or
maybe .bashrc) and do not resume until I issue a Control-C to kill the
offending process.
I did not set that DISPLAY myself; some daemon did it, I suppose.

If prior to the update you were using one of the Xquartz releases,
then install the 2.3.1 release.
If you were using only the 10.5.x provided X11, then if you updated
via software update, then try downloading the combo updater and
applying it.
