In principle (when you don't use any 10.6 API) it shouldn't matter whether you build on a machine running 10.5 or 10.6. I do hope that you can develop on 10.6 though, because you should test your app on all systems for which you develop. Moreover, 10.5 doesn't support newer Xcode versions, so you have a bigger chance to run into a bug in Xcode (for instance for cross-project support). So I would recommend to build on the newest OS version if you can, but it's not necessary.
