There are actually two things to consider here.
The first is when creating a Hexachrome profile you need to use a composite TIFF + alpha channels to ensure the plates are correctly recognized by ColorBurst. Colorburst only accepts CMYK + alpha channels and not DCS files for pre-seperated files. You need to have the alpha channels inverted (black) for the ColorBurst to properly process them. Onyx and EFI an most others accept DCS files in addition to TIFFs.
The original question was if you could color manage CMYK files with PMS bump plates to a Hexachrome setup. If you print (postscript) or PDF that has the bump plates showing up separately from the CMYK plates then Colorburst will color manage those spots to the full gamut of the Roland Hexachrome ICC profile AND manage the CMYK objects to the Roland Hexachrome profile too.

Yup. ColorBurst doesn't like separated postscript (EPS or DCS) you need to send TIFFs as CMYK + inverted alpha channels. It's possible that the PDF data wasn't parsed correctly knowing that DCS and EPS files don't parse.
I haven't tried the PDF shtick with ColorBurst, I just chugged away with the composite TIFFs.

So in summary, composite TIFFs w/ alpha channels for Hexachrome profiling, and postscript/pdf for CMYK + spot color matching to Hexachrome profile.
