It seems logical to me that you should build your sampler as an MusicDevice Audio Unit because then you can take advantage of all the different hosting environments available (so you don't have to build your own sequencer for example) - also who knows what you'll want to do with it in the future - like adding effects etc.

And if you just want to build your own host then the AUGraph stuff is very easy and tidy to work with.
