This worked just now when I tried with Instruments from my Xcode 3.2.1 install on Snow Leopard 10.6.2:

	* Launch Instruments
	* Switch to some other app
	* Hit the default Ctrl+Opt+Esc global keyboard shortcut
	* A bezel declaring that sampling had started was displayed
	* Hit the same keyboard shortcut to stop sampling
	* A bezel declaring that sampling had stopped was displayed
	* The Instrument dock icon shows a badge with the number of available samplings
	* Right-click the Instruments dock icon to select the sample
